Subcellular Distribution in Rat Cerebral-cortex Slices

چکیده

1. Rat cerebral-cortex slices were incubated with 32pl, acetylcholine and eserine for periods of 10min and 2h. The specific radioactivity of phosphatidylinositol was elevated during these treatments by 36 and 106% respectively. 2. The specific radioactivities of the phosphatidylinositol in different cell structures were determined after subcellular fractionation. They were highest in the nuclear, microsomal and synaptic-vesicle fractions and lowest in myelin, both in the controls and in the acetylcholine-treated slices. 3. The stimulated labelling of phosphatidylinositol was relatively evenly distributed: no subcellular fraction showed a stimulation markedly higher than that in the homogenate. 4. Studies of the distributions and activities of marker enzymes indicated that the subcellular fractionation achieved was similar to that with fresh tissue. 5. The results are discussed in relation to the previous report that the stimulation is observed throughout the neuronal cell-bodies and in relation to the hypothesis that the labelled phosphatidylinositol produced by stimulation is a component of an acetylcholine-receptor proteolipid localized in the synaptic junction.

Subcellular distribution of proteins synthesized in slices of rat cerebral cortex.

The incorporation of L-[4,5-3H]leucine into slices of rat cerebral cortex has been measured, with emphasis on the time course of the distribution of label in subcellular fractions prepared by zonal isopycnic centrifugation in continuous sucrose gradients. Rapidly labeled proteins are found in both the soluble and membranous components of the nerve end particles. Except for a minor part of the m...

Effects of visual deprivation on epileptic activity in mature rat visual cortex

Effects of visual deprivation on the induction of epileptiform activity were studied in layer II/III of mature rat primary visual cortex. Field potentials were evoked by stimulation of layer IV in slices from control and dark-reared (OR) rats. Picrotoxin (PTX)-induced epileptic activity was characterized by spontaneous and evoked epileptic field potentials (EFPs). The results showed that OR s...

The Subcellular Localization of Cerebral Phosphoproteins Sensitive to Electrical Stimulation.

1. On incubating cerebral-cortex slices at 37 degrees in an oxygenated medium marked changes resulted in the subcellular distribution of proteins and phosphoproteins in the tissue. The protein content of the nuclear fraction more than doubled, whereas the yields of microsomal and supernatant proteins were both markedly decreased. The amount of phosphoprotein/mg. of protein decreased in the micr...

Cell Type–Specific Thalamic Innervation in a Column of Rat Vibrissal Cortex

This is the concluding article in a series of 3 studies that investigate the anatomical determinants of thalamocortical (TC) input to excitatory neurons in a cortical column of rat primary somatosensory cortex (S1). We used viral synaptophysin-enhanced green fluorescent protein expression in thalamic neurons and reconstructions of biocytin-labeled cortical neurons in TC slices to quantify the n...

The distribution of gangliosides in subcellular fractions of guinea-pig cerebral cortex.

Gangliosides are acidic, acid-sensitive, nondialysable, water-soluble lipids containing Nacetylneuraminic acid, hexosamine, hexoses and ceramides which in the brain occur mainly in grey matter. The aim of the present investigation was to study the localization of gangliosides in the cerebral cortex by following their distribution in subcellular fractions, neuraminic acid content being used as m...
